Основные труды — в области нейрогистологии современных беспозвоночных, сравнительной морфологии и палеоневрологии ископаемых животных. Главный редактор «Основ палеонтологии» (т. 1-15). В 1967 году награждён Ленинской премией посмертно. Именем Орлова назван палеонтологический музей в Москве.
